A rapid survey of Asp isomers in aged proteins
نویسندگان
چکیده
Background: Asp-isomers in lens crystallins are one of the triggers of cataracts. Results: Multiple highly isomeric Asp sites in insoluble crystallins from cataract lenses were identified by LC-MS using the corresponding synthetic peptides as standards. Conclusion: Asp-isomers induce insolublization of crystallin, leading to cataracts. Significance: This study opens up a new field of protein biochemistry in age-related diseases.

Kinetics of Isomerization and Inversion of Aspartate 58 of αA-Crystallin Peptide Mimics under Physiological Conditions
Although proteins consist exclusively of L-amino acids, we have reported that aspartyl (Asp) 58 and Asp 151 residues of αA-crystallin of eye lenses from elderly cataract donors are highly inverted and isomerized to D-β, D-α and L-β-Asp residues through succinimide intermediates. Of these Asp isomers, large amounts of D-β- and L-β-isomers are present but the amount of D-α-isomer is not significa...
متن کاملA Rapid and Eco-friendly Synthesis of 5(6)-Carboxy-Rhodamine 110 Isomers
5(6)-Carboxy-rhodamine 110 isomers were rapidly synthesized by condensation of 3-aminophenol and trimellitic anhydride using microwave irradiation as heating method. After esterification of the two isomers, separation and hydrolyzation provided target products,5-carboxy-rhodamine 110 and 6-carboxy-rhodamine 110, in high yields. The results shows microwave irradiation can effectively reduce ...
متن کاملD-β-aspartyl residue exhibiting uncommon high resistance to spontaneous peptide bond cleavage
Although L-amino acids were selected as main constituents of peptides and proteins during chemical evolution, D-aspartyl (Asp) residue is found in a variety of living tissues. In particular, D-β-Asp is thought to be stable than any other Asp isomers, and this could be a reason for gradual accumulation in abnormal proteins and peptides to modify their structures and functions. It is predicted th...

متن کاملWavelength region selection and spectrophotometric simultaneous determination of naphthol isomers based on net analyte signal
Naphthol isomers were simultaneously and spectrophotometrically determined in wastewater, using a model based on net analyte signal (NAS). The calibration method used is a variation of the original hybrid linear analysis method as proposed by Goicoechea and Olivieri (HLA/GO). Owing to spectral interferences, the simultaneous determination of mixtures of naphthol isomers, using a spectrophotomet...
